Varying flow in linear dichroism cells

چکیده
The usefulness of linear dichroism (LD) in structure identification depends strongly on the orientation of the molecule in the linear dichroism cell. Hence, the fluid flow inside the linear dichroism cell which performs this act of orientation is of great importance. As such, this paper records the steps taken to identify the varying flow states in the cell, with changing rotation speed, and the resulting effects on the orientation of the molecule, as deduced from the LD spectra obtained. The previously known stiffening effects of ethidium bromide (EB) on DNA are reproduced to a certain extent, the change appearing for high concentrations of EB, but, significantly, at the same point at which Taylor vortices appear in the flow. The inability to scale up the results obtained from the micro-volume LD cell to match those from the larger cell suggest a dependence of the orientation on the flow profile in the cell, which are different in the two cells.

Micro-volume couette flow sample orientation for absorbance and fluorescence linear dichroism.
Linear dichroism (LD) can be used to study the alignment of absorbing chromophores within long molecules. In particular, Couette flow LD has been used to good effect in probing ligand binding to DNA and to fibrous proteins. This technique has been previously limited by large sample requirements. Here we report the design and application of a new micro-volume Couette flow cell that significantly...
متن کاملBreaking the 200 nm limit for routine flow linear dichroism measurements using UV synchrotron radiation.
The first synchrotron radiation flow linear dichroism spectra are reported. High-quality spectral data can be collected from 450 nm down to 180 nm in contrast to the practical cutoff of approximately 200 nm on benchtop instruments. State-of-the-art microvolume capillary Couette flow linear dichroism was successfully ported to a synchrotron radiation source. The sample volume required is < 50 mi...
متن کاملTime-Resolved Linear Dichroism Measurements of Carbonmonoxy Myoglobin as a Probe of the Microviscosity in Crowded Environments.
The distribution of viscosities in living cells is heterogeneous because of the different sizes and natures of macromolecular components. When thinking about protein folding/function processes in such an environment, the relevant (micro)viscosity at the micrometer length scale is necessarily distinguished from the bulk (macro)viscosity. The concentration dependencies of microviscosities are det...
متن کاملProtein fiber linear dichroism for structure determination and kinetics in a low-volume, low-wavelength couette flow cell.
High-resolution structure determination of soluble globular proteins relies heavily on x-ray crystallography techniques. Such an approach is often ineffective for investigations into the structure of fibrous proteins as these proteins generally do not crystallize. Thus investigations into fibrous protein structure have relied on less direct methods such as x-ray fiber diffraction and circular d...
متن کاملStructural transitions of chromatin at low salt concentrations: a flow linear dichroism study.
We have studied the structure of nuclease-solubilized chromatin from Ehrlich ascites cells by flow linear dichroism (LD) using the anisotropic absorption of the DNA bases and of two intercalated dyes, ethidium bromide and methylene blue. It is confirmed that intercalation occurs preferentially in the linker part of the chromatin fiber, at binding ratios (dye/base) below 0.020. Using this inform...
